1. What It Is & How It Works

Saridon is a fixed‑dose combination analgesic pairing:

• Paracetamol (acetaminophen) – analgesic and fever reducer, slower onset but longer duration

• Propyphenazone – pyrazolone analgesic with faster onset

• Caffeine – mild stimulant that enhances pain relief, narrows cerebral blood vessels, and reduces drowsiness

This combination boosts propyphenazone’s peak plasma levels by ~40% and prolongs its effect (~77 min half‑life), offering quick and sustained relief.

Standard Indian formulation (Saridon Head & Body) contains:

• Paracetamol 250 mg

• Propyphenazone 150 mg

• Caffeine (anhydrous) 50 mg

2. Key Uses & Benefits

Used to relieve short‑term pain from:

• Primary headaches (tension, mild migraine), including back‑of‑head or diffuse pain

• Toothache, muscle ache, or backache

• Mild fever, menstrual cramps when body pain predominates

• Cold/flu discomforts with body ache and headache

3. Dosage (Adults 12+) & Onset

• Wait at least 4 hours between doses

• Symptoms usually ease within 30–60 min and return after ~4–6 hours

• If pain persists > 3–5 days, consult a physician rather than continuing OTC use

Children under 12: Not recommended
Pregnancy / breastfeeding: Avoid unless advised by a doctor

4. Possible Side Effects & Safety Considerations

Common, usually mild (1–10%):

• Nausea, indigestion, abdominal discomfort, bloating

• Mild skin rash or itching, headache, dizziness (rare)

• Dry mouth, restlessness due to caffeine  

Rare but serious (estimate < 0.01%):

• Allergic reactions: angioedema, severe rash, difficulty breathing

• Hematological toxicity: thrombocytopenia, leukopenia, agranulocytosis, pancytopenia

  • Mostly linked to Propyphenazone; propyphenazone has been associated in isolated instances with Kounis syndrome (allergic heart issues) and blood dyscrasias

Liver warning: overdose or prolonged intake—especially with daily alcohol—can cause acute hepatic injury (paracetamol toxicity). Monitor warning signs: nausea, jaundice, dark urine.

Stimulant caution: caffeine may cause restlessness, insomnia, rapid heart rate, especially if other caffeine-containing foods/drinks are consumed.

5. When to Avoid or Use With Caution

Not suitable for:

• Allergy to paracetamol, propyphenazone, NSAIDs, aspirin, or caffeine

• Children under 12 (or in some countries under 6)

• Pregnant or breastfeeding women, unless prescribed

• Bone marrow disorders, G6PD deficiency, chronic agranulocytosis anemia

• Bleeding/ulcer disease, hepatic porphyria, chronic alcoholism

Use with caution in: liver or kidney disease, chronic malnutrition, asthma (some NSAID-sensitive types), seizure history, or cardiovascular disease.

6. Drug & Food Interactions

Avoid if possible: other medicines containing paracetamol (acetaminophen), caffeine (e.g. colas, coffee), or NSAIDs.
Liver-inducing drugs — such as barbiturates, phenytoin, rifampicin, carbamazepine — can increase risk of toxic paracetamol metabolites. Alcohol also raises liver risk.
Zidovudine, used in HIV, may add to hematologic toxicity.
Caffeine may reduce absorption or cancellation of sedatives; avoid doubling stimulant effects.

 Special Notes & History

• First introduced in 1933 (then under Roche), reformulated in 1981 to exclude phenacetin; originally banned in several countries including Sri Lanka, Malaysia, and Thailand due to safety concerns.

• In September 2018, India’s regulator banned 327 FDCs including Saridon; the Supreme Court stayed the ban pending reassessment of safety data, allowing its continued sale subject to compliance.

• Current formulation in India is marketed by Bayer Pharmaceuticals (via Piramal Enterprises) and approved for short-term OTC use.

8. How to Use & Storage Tips

• Swallow with water; do not crush or chew

• Preferably taken after food to avoid mild gastritis

• Limit additional caffeine intake on the same day

• Store below 25 °C, in a dry place, out of reach of children

• Do not exceed 5 days’ continuous use without medical review
